چکیده
Abstract Introduction Erectile dysfunction is associated with increased contraction and decreased relaxation of the corpus cavernosum (CC). Excessive reactive oxygen species produced by mitochondria play a key role in aging- diabetes-induced erectile dysfunction. cavernosum. Mitochondrial deterioration part normal physiological aging, it plays pathogenesis age-related disorders producing (ROS). It also known that excessive ROS Objective We aimed to test hypothesis vitro inhibition mitochondrial oxidative stress improves reactivity CC. Methods To this hypothesis, CC young (3 4-month-old) old (10 14-month-old) male lean diabetic (dbdb-/-) mice were mounted myographs. Concentration-response curves acetylcholine (ACh 1 nM – 30 uM) induced electrical field stimulation (EFS 1, 2, 4, 8 16 Hz) obtained. All repeated after minutes incubation antioxidant Mito-Q (100 nM). Results Results: Relaxation ACh was significantly reduced (Lean: 87 ± 8% vs dbdb-/-: 60 6%). Similarly, aged presented impaired (Lean old: 54 6%) while displayed further decrease relaxing response (35 After Mito-Q, improved (62 3%) (44 1%). Relaxing responses EFS obese compared animals. However, no improvement seen Mito-Q. Conclusions Impairment endothelium-dependent aging diabetes production species. The use mitochondrial-targeted at least both mice. Disclosure No

Nebivolol, but not metoprolol, improves endothelial function of the corpus cavernosum in apolipoprotein e-knockout mice.
To determine the effects and underlying mechanisms of treatment with the beta-receptor blockers nebivolol and metoprolol on penile endothelial function in apolipoprotein E (ApoE)-/- mice, wild-type (WT) and ApoE-/- mice were fed with a cholesterol-rich diet for 7 weeks. ApoE-/- mice were treated with nebivolol (10 mg/kg/day) or metoprolol (90 mg/kg/day). Endothelial function of aortic and corpo...
متن کاملMitochondria-Targeted Antioxidant SkQ1 Improves Dermal Wound Healing in Genetically Diabetic Mice
Oxidative stress is widely recognized as an important factor in the delayed wound healing in diabetes. However, the role of mitochondrial reactive oxygen species in this process is unknown. It was assumed that mitochondrial reactive oxygen species are involved in many wound-healing processes in both diabetic humans and animals. We have applied the mitochondria-targeted antioxidant 10-(6'-plasto...

PURPOSE To describe a technique for en bloc harvesting of the corpus cavernosum, cavernous artery and urethra from transplant organ donors and contraction-relaxation experiments with corpus cavernosum smooth muscle. MATERIALS AND METHODS The corpus cavernosum was dissected to the point of attachment with the crus penis. A 3 cm segment (corpus cavernosum and urethra) was isolated and placed in...

Main outcome measures Cumulative concentration-response curves were constructed for acetylcholine (ACh), sodium nitroprusside (SNP), 5-cyclopropyl-2-[1-(2-fluorobenzyl)-1H-pyrazolo [3,4-b]pyridine-3-yl]pyrimidin-4-ylamine (BAY 41-2272), and tadalafil in CCC precontracted with phenylephrine. Relaxation induced by electrical field stimulation (EFS) was also done in the absence and presence of Nw ...
